Isolation and Characterization of Thiomicrospira sp. V2501 Strain V2501 was isolated on December 15, 2009, from groundwater obtained via borehole 09-V250-M02, located 250 m below ground level at the Horonobe Underground Research Center. It is capable of chemolithoautotrophic growth, utilizing carbon dioxide and thiosulfate as substrates. 2. Genome Sequencing and Analysis The complete genome of strain V2501 was obtained using the PacBio Sequel IIe system. ・Genome size : 2,240,851 bp ・GC content : 45.1 mol% ・Identified genes : 2,135 protein-coding genes, 9 RNA genes, and 43 tRNA genes 3. Phylogenetic Analysis Phylogenomic analysis using TYGS revealed that strain V2501 is closely related to Thiomicrospira pelophila DSM 1534T, with an OrthoANIu value of 83.2%, which falls below the species-level cutoff of 95% ± 0.5%. --- Conclusion The complete genome sequencing of Thiomicrospira sp. V2501 provides valuable insights into the metabolic mechanisms and ecological roles of subsurface microbes. The genomic data will facilitate further studies on the chemolithoautotrophic pathways of strain V2501 and its contributions to carbon and sulfur cycling in deep subsurface environments.
